Well, seeing as DQV was my all-time fave you knew I'd have to chime in at some point...
The fact that you get 4 characters out in your party instead of just 3...the wife makes a bigger difference here, and I think Bianca is the best choice...followed by Deborah...she's funny, and she gets to use weapons the others don't.
As to the T&T...SAVE THOSE TICKETS!!
Save it for the fourth T&T board.
One of the prizes on the fourth board, for completing is, it a Golden T&T Ticket...that gives you hitherto unlimited access to the T&T Boards.
The main thing you can use T&T Boards for is to get into fights with "forever gone" monsters to complete your book of beasts, even though the cheap bastards gave no prize for completing it in this game.
I have gotten the 45,000 gold weapon, Staff of Resurrection, just lying around in those boards...usually the fifth board.
It is NOT worth going thru the first 3 T&T boards over and over again. 4 and 5 are a different matter.
Especially because of the Treasure Rooms in those boards...you want to get all the treasure. There's some good stuff to be had in the treasure rooms!

: Cosmetic difference. Your children's stats,
: abilities, and equipment always stay the
: same.
:
: Not to mention that, at least in the SNES
: original, the game *really* wants you to
: marry Bianca. If you don't, the game
: *really* heaps on the guilt. Bianca's father
: dies, Bianca is forced to work as a barmaid,
: and Flora's (Nera, in the DS version)
: boyfriend, who risked his life in the
: volcano trying to get the ring, is left out
: in the cold. Moreover, I've heard Flora/Nera
: starts at level 1, and even levelled up, is
: nigh-useless in battle. Seriously, why did
: they *bother*?
